What are the differences in the harvesting protocol when recovering intact organoids versus organoid fragment (dissociated organoid)? Does incubation time impact organoid integrity, or is it primarily influenced by mechanical factors such as the type of pipette used?

The VitroGel® Organoid Recovery Solution was designed to harvest organoids intact. The medium doesn’t support the dissociation of the organoid into single cells. If dissociating the organoid into single cells is needed, you can add the dissociated solution after harvesting the organoid from the VitroGel® system or mix the dissociated solution with VitroGel® Organoid Recovery Solution at a 1:1 ratio while harvesting the organoid from the hydrogel system. In terms of the incubation time and mechanical factor, both will impact the organoid harvesting results, inceasing the incubation time can help to dissolve the hydrogel, and adding the mechanical force such as a pipette to mix the gel-organoid-recovery solution mixture will help to accelerate the cell harvesting and break the organoid into small fragments.
